TLR3(又名CD283)是一個胞內(nèi)受體,可識別與病毒感染相關(guān)的雙鏈RNA(dsRNA)分子模式。其中一個TLR3的配體,Poly(I:C),是人工合成的dsRNA類似物。TLR3信號通路不依賴于MyD88,而是通過接頭蛋白TRIF(包含TIR結(jié)構(gòu)域的接頭蛋白,可誘導(dǎo)IFN-β產(chǎn)生)發(fā)揮作用。IRF3(干擾素調(diào)節(jié)因子3)被磷酸化后導(dǎo)致IFN-β(β干擾素)產(chǎn)生,進(jìn)一步活化ISRE(干擾素激活反應(yīng)元件)。另外,TLR3信號通路的激活也可導(dǎo)致AP-1和NF-kB等轉(zhuǎn)錄因子入核,促進(jìn)一些炎癥因子的釋放。
