轉(zhuǎn)錄因子分析?(Transcription?Factor?Assays)

Cayman研發(fā)的轉(zhuǎn)錄因子分析技術(shù),采用非放射性、高靈敏度的方法,檢測(cè)細(xì)胞核提取物中轉(zhuǎn)錄因子與DNA的特異性結(jié)合活性。該技術(shù)取代傳統(tǒng)繁雜的EMSA方法,用96孔板形式來(lái)分析轉(zhuǎn)錄因子與DNA的結(jié)合。主要技術(shù)原理為,首先將特異性dsDNA序列(含應(yīng)答元件)包被固定于96孔板中,再將含轉(zhuǎn)錄因子的核提取物加入板孔中孵育,轉(zhuǎn)錄因子與包被的DNA序列特異性結(jié)合,然后用轉(zhuǎn)錄因子特異性的一抗孵育,最后加入相應(yīng)的二抗-HRP結(jié)合物,顯色并于450nm讀數(shù)。
