8月15日,清華大學生物醫(yī)學交叉研究院教授、北京生命科學研究所副所長邵峰院士團隊在Nature期刊(IF=41.577)上在線發(fā)表了題為”Alpha-kinase 1 is a cytosolic innate immune receptor for bacterial ADP-heptose”的研究論文,首次發(fā)現并證明哺乳動物細胞質內的一個新的激酶分子ALPK1(alpha-kinase 1)可以直接和特異性識別細菌脂多糖LPS(俗稱內毒素)合成的前體糖分子ADP-heptose(二磷酸腺苷庚糖),進而激活NF-κB通路介導的天然免疫炎癥反應。這項研究揭示了宿主細胞內的全新激酶受體蛋白ALPK1可以識別細菌來源的ADP-heptose,代表了一種全新的、保守存在于脊椎動物中的抗細菌天然免疫模式識別通路,這一發(fā)現也豐富和改變了人們對LPS相關分子誘導炎癥反應機制的認識,為開發(fā)新的免疫調節(jié)劑和疫苗佐劑提供了新的概念和方法。?

本項研究發(fā)現,給小鼠直接注射ADP-heptose可以引起強烈的ALPK1依賴的炎癥反應,而注射HBP則與注射生理鹽水一樣不能引起任何炎癥反應。進一步在野生型和Alpk1基因敲除的小鼠中進行了洋蔥伯克霍爾德菌(Burkholderia cenocepacia)的肺部感染實驗,發(fā)現Alpk1敲除的小鼠肺部炎癥反應程度更輕,同時細菌數量顯著高于野生型小鼠。這一結果證實了ADP-heptose-ALPK1天然免疫信號通路在抵御細菌感染過程中發(fā)揮重要作用。該實驗中使用了欣博盛Mouse IP-10/CXCL10 ELISA kit(小鼠干擾素誘導蛋白10試劑盒)來檢測小鼠氣囊液上清的IP-10濃度。?

8月1 日,上海藥物所藥物制劑中心李亞平課題組在國際權威期刊Advanced Materials(IF=21.95)上發(fā)表了題為”Binary Cooperative Prodrug Nanoparticles Improve Immunotherapy by Synergistically Modulating Immune Tumor Microenvironment”的文章。此項研究創(chuàng)新性設計構建了一種智能前藥納米粒 (Binary cooperative prodrug nanoparticles, BCPN),實現二元協同、雙管齊下高效免疫治療腫瘤。研究發(fā)現:BCPN可以把化療藥物奧沙利鉑的前藥分子和IDO-1酶抑制劑NLG919的二聚體高效共遞送到腫瘤組織,然后在腫瘤細胞還原環(huán)境中,前藥分子被還原激活釋放活性奧沙利鉑和NLG919單體。奧沙利鉑可刺激腫瘤細胞發(fā)生免疫性細胞死亡,提高腫瘤組織免疫原性,誘導機體對腫瘤細胞的特異性免疫反應,增加腫瘤組織內CTL的浸潤,促使“冷瘤”轉變?yōu)椤盁崃觥?。NLG919可有效抑制IDO-1酶活性,緩解IDO-1酶對CTL的抑制作用,克服腫瘤抑制微環(huán)境。BCPN在4T1乳腺癌和CT26結腸癌的小鼠模型中均表現出良好的抗腫瘤轉移和復發(fā)效果,該工作對發(fā)展安全高效的腫瘤免疫治療具有重要參考價值。?
