人體內(nèi)的細(xì)菌數(shù)量是人體細(xì)胞的10倍。這些微生物群廣泛存在腸道上皮和粘膜細(xì)胞表面,對維持機(jī)體健康起著非常重要的作用。如果正常的共生微生物群(腸道菌群)的生態(tài)平衡發(fā)生紊亂,將嚴(yán)重影響人體健康,引發(fā)風(fēng)濕性關(guān)節(jié)炎、炎癥性腸病、多發(fā)性硬化癥及糖尿病等多種疾病。除了能抵御有害病菌造成的物理及化學(xué)的傷害,腸道菌群在維持免疫功能(Th17,Treg及固有淋巴細(xì)胞)方面也發(fā)揮著重要的作用。
腸道免疫系統(tǒng)能區(qū)分共生菌和病原菌,對共生菌產(chǎn)生免疫耐受,而對病原菌及其他抗原物質(zhì)產(chǎn)生免疫反應(yīng)。Th17細(xì)胞是能產(chǎn)生IL-17的CD4+T淋巴細(xì)胞亞群,Th17的產(chǎn)生依賴于腸道共生菌的存在,并成為腸道免疫屏障的重要組成部分。它的適量存在有助于維持腸道免疫功能,而Th17的過度誘導(dǎo)則會(huì)導(dǎo)致腸道免疫功能破壞。此外,在腸道免疫系統(tǒng)中對共生菌耐受的CD4+CD25+Treg細(xì)胞及其產(chǎn)生的一致性細(xì)胞因子IL-10和TGF-β也發(fā)揮著重要作用,Th17細(xì)胞的分化發(fā)育又與Treg細(xì)胞分化發(fā)育呈抑制性關(guān)系。因此,Th17和Treg細(xì)胞是連接著腸道菌群和腸道免疫功能的關(guān)鍵細(xì)胞亞群。
